Because melanoma cells were reported to\nexpress a large amount of SPARC, and because SPARC is a secreted protein, at first, secreted\nSPARC protein was quantified using ELISA in the sera from 109 melanoma patients, 5\npatients with large congenital melanocytic nevus, 61 age-matched healthy donors and 13\ndisease-free patients after undergoing a surgical removal. We also quantified two other known\ntumor markers for melanoma, GPC3 and 5-S-cysteinyldopa (5-S-CD), in the same serum\nsamples, and compared these markers for their diagnostic value. Because of the similarities in\nthe binding peptide motifs shared between H2-Kd and HLA-A24 (A*2402), 4 peptides having\nthese motifs were synthesized. Then, we investigated whether the H2-Kd-restricted SPARC\npeptides could induce SPARC reactive cytotoxic T lymphocytes (CTLs) or not.
