| 内容記述 | 胃癌において腹膜播腫は再発形式として最も多く難治性の病態である。アデノウイルスを用いた遺伝子治療は、この病態に対する有用な治療法の一つになりうる可能性がある。しかし、アデノウイルスベクターには臨床応用するにあたり、いくつかの問題点が存在する。その1 つとして、アデノウイルスベクターの容量依存的毒性が挙げられる。それを克服するために、我々は従来のSuicide gene therapy とOncolytic gene therapy を組み合わせたOncolytic suicide gene therapy という新しい遺伝子治療システムを考案した。このシステムではCEA 産生細胞選択的にこれら2 つの従来法の相乗効果が発揮できるように 設計されている。我々は、このシステムを用いて抗腫瘍効果を保ちながらウイルスベクター総投与量の減量が可能かどうかを、胃癌腹膜播腫モデルマウスを用いて検討した。 |

| 内容記述 | The peritoneal dissemination of gastric cancer is often refractory to systemic therapies. Although adenoviral gene therapy has been reported to be a potentially useful therapeutic modality, the adenovirus itself has a dose-limiting toxicity. In this study, a novel system was constructed using adenoviral oncolytic suicide gene therapy targeting carcinoembryonic antigen (CEA), and assessed the therapeutic effect and possibility to reduce the total viral dose while still preserving the antitumor effect in peritoneal dissemination models of gastric cancer in mice. |
